Fine cytochemical localization of lysosomal organelles in phagocytic cells of bivalve molluscs. ArchiMer

Bivalve molluscs possess several types of hemolymph cells (hemocytes), involved in internal defense and other physiological functions. Studies in pathological and experimental conditions have shown their micro- and macrophagic ability. However, the mechanisms of intracellular digestion are incompletely known in these organisms. The lysosomal apparatus of hemocytes was therefore investigated in several species by hydrolase localization with the electron microscope. The cytoplasmic granules of granulocytes displayed a strong acid beta -glycerophosphatase activity. Previous observations had shown that these granules may contain phagocytized material. Hemócitos fagocitários em larvas de Diatraea saccharalis (Fabricius) (Lepidoptera, Pyralidae) Zoologia

The ultrastructural observation of the hemocytes of D. saccharalis larvae showed different hemocyte tvpes in various stage of interaction with microorganisms, which were interpreted as bacteria. The microorganisms were observed either free in the hemolymph or into the insect hemocytes. The bacteria were detected mostly into lhe granulocytes and plasmatocytes: the spherule cells and the oenocytoids also presented intracytoplasmic microorganisms, but less frequently. The fagocytic capacity of the different hemocyte tvpes is discussed.

Bovine S protein (vitronectin) increases phagocytosis of Streptococcus dysgalactiae Rev. Microbiol.

The effects of bovine S protein (vitronectin) on phagocytosis of Streptococcus dysgalactiae strains isolated from cattle with mastitis were investigated. Phagocytized streptococci were determined by a fluorometric microassay using glass adherent polymorphonuclear neutrophils (PMN). Preincubation of S. dysgalactiae with bovine S protein significantly increased their phagocytosis by PMN. Bovine S protein had no effect on phagocytic killing of non-S protein binding S. pyogenes cultures. Enzymatic digestion of the bovine S protein binding sites on S. dysgalactiae with pronase resulted in a significative reduction of the effects of S protein on phagocytosis. Fish present an efficient and developed immunologic response and in the case of Teleosts, with some similarities to the higher vertebrates. Role of previous stimulation on the neutrophil migration protects against the lethal effect of Salmonella typhimurium Braz. J. Vet. Res. Anim. Sci.

The intraperitoneal inoculation of 3.8 x 10(9) CFU of alive Salmonella typhimurium caused 100% mortality in rats. In these animals a small number of neutrophils migrated to the peritoneal cavity. The intraperitoneal inoculation of the animals with thioglycollate (Tg, 90 mg), carrageenin (Cg, 500 mu g), or killed S. typhimurium (KS, 1.9 x 10(9) CFU) 24 hours before challenge with alive S. typhimurium caused a significant neutrophil migration to the peritoneal cavity (determined 6 hours after challenge) and protected the animals against the lethal effect of the bacteria at a rate of 50%, 83% and 80%, respectively. However, similar treatment of the animals with brain heart infusion broth (BHI, S. typhimurium grout medium) protected the animals by only 23%....

The uptake of lipids and lipoprotein particles by macrophages undergoes phagocytic activation and the formation of foam cells are key events in atherosclerosis. In this study we determined how intact high density lipoproteins (HDL) and apolipoproteins-HDL (removal of the lipid component from HDL, i.e., apoHDL) influence the phagocytosis of zymosan by mouse peritoneal macrophages. Zymosan particles preincubated together with lipoproteins or alone (control) were incubated with the macrophages. Bonamia ostreae is the causative agent of hemocyte disease. It has been detected first during mortalities occured in Franc in 1979. Bonamiosis spread rapidly among oysters farming in France and Europ. Another species, Bonamia sp. has been detected in New Zealand and Australia. These paasites were both included in the "microcell" group. This group contains 2 others parasites, Mikrocytos mackini affecting Crassostrea gigas in British Columbia (Canada) and M. roughleyi affecing Saccostrea glomerata in Australia. Several studies show the ability of macrophages to remove particles injected into the bloodstream. This function seems to be increased in the presence of acute renal failure. The objective of the present study was to assess the phagocytic function of the main organs (spleen, liver and lung) of the mononuclear phagocytic system in renal and postrenal failures. Fifteen rats (250-350 g) were divided into three groups (N = 5): group I - control; group II - ligature of both ureters, and group III - bilateral nephrectomy. Role of Tamm-Horsfall protein in the binding and in vivo phagocytosis of type 1 fimbriated Escherichia coli by mouse peritoneal macrophages BJMBR

Tamm-Horsfall glycoprotein (THP) contains manno-oligosaccharides that are recognized by type 1 fimbriae (F1) of Escherichia coli. In the present study, we examined the in vivo phagocytic activity of mouse peritoneal macrophages after treatment of bacteria with THP. At low THP concentrations (12.5 µg/ml and 50 µg/ml) no significant difference was observed in the phagocytosis of E. coli F1+. However, at high THP concentrations (500 µg/ml and 1250 µg/ml) we obtained a reduction of bacterial phagocytosis by mouse peritoneal macrophages.

Previous studies have demonstrated that some components of the leukocyte cell membrane, CR3 (Mac-1, CD11b/CD18) and p150/95, are able to bind to denatured proteins. Thus, it is of interest to know which effector functions of these cells can be triggered by these receptors when they interact with particles or surfaces covered with denatured proteins. In the present study we analyzed their possible role as mediators of phagocytosis of red cells covered with denatured bovine serum albumin (BSA) by mouse peritoneal macrophages. Treatment of serum with supernatants from cultures of Candida albicans reduces its serum-dependent phagocytosis BJM

Candida albicans is a potent activator of the complement system, and heat labile opsonins produced by activation of C3 (C3b and iC3b) enhance phagocytosis of C. albicans mediated by complement receptors. In this study we treated mouse serum with supernatants from cultures of a protease producer strain of C. albicans and evaluated the ability of this serum to enhance phagocytosis of C. albicans. Cell-free supernatants from cultures of C. albicans were concentrated 5 fold and added to mouse serum for 30 min at 37ºC, before using this serum for opsonization of glutaraldehyde-fixed yeast cells. The impact of diets upon the fatty acid composition of haemocyte polar lipids and consequently upon immune parameters has been tested in the oyster Crassostrea gigas and the clam Ruditapes philippinarum. The defense reactions against biological (Histoplasma capsulatum and Escherichia coli) and non-biological materials (China ink and nylon thread) were tested in vivo in third instar larvae of Dermatobia hominis. The cellular defense performed by larval hemocytes was observed under electron microscopy. China ink particles were phagocytosed by granular cells 5 h after injection. Evaluation of possible failure of the mononuclear phagocyte system after total splenectomy in rats BABT

Young and adult Wistar rats were submitted to total splenectomy and compared to animals not submitted to any surgical manipulation in order to evaluate the phagocytic function of spleen. The animals were infected with Escherichia coli labeled with technetium-99m and killed 20 minutes later. Liver, lung, spleen and a blood clot sample were taken. No significant differences were found in the percentage of bacterial radioactivity uptake in mononuclear phagocyte system (MPS) organs in young and adult splenectomized rats. In the context of Pacific oyster C. gigas summer mortality, the present study demonstrated that the reproductive effort reduced not only the energetic parameters (carbohydrates and CEA) but also the immune parameters (concentration, phagocytosis and adhesive capacity of hemocytes). Increasing food supply from 4% to 12% (of oyster dry weight) did not counteract this phenomena as most of the additional energy was dedicated to the gametogenesis processes. Effects of extracellular products from the pathogenic Vibrio aestuarianus strain 01/32 on lethality and cellular immune responses of the oyster Crassostrea gigas ArchiMer

Vibrio aestuarianus strain 01/32 was previously shown to be pathogenic to Crassostrea gigas juveniles. To investigate virulence mechanisms of this pathogen, we studied the toxicity to oysters of its extracellular products (ECPs). ECPs displayed lethality to animals, with a LD50 value of 3.3 mu g/g body weight. To determine the oyster cellular immune responses induced by these ECPs, we further examined in vitro their effects on C. gigas hemocytes, using flow cytometric-based hemocyte assays. The aim of this study was to test the effect of food quantity on energy storage and defense capacities of oysters during a reproductive cycle. One-year-old Crassostrea gigas oysters were fed two different dietary rations (4% and 12% of oyster dry weight in algal dry weight per day) in controlled experimental conditions over an annual cycle.
